ルート66はシカゴからロサンゼルスに至る全長3,755km（2,347マイル）の大陸横断ハイウェイ66号線道路のことで、イリノイ、ミズーリ、カンザス、オクラホマ、テキサス、ニューメキシコ、アリゾナ、カリフォルニアの8州にまたがる「母なる道（Mother road）」として1926年に開通した。
戦後ハイウエーの発達するにつれその役割を失い、分断されて幹線地図からも消えた。
現在は地方道として残っているだけ。
そのルート66を、トッドとバズの青年二人がシボレー・コルベットのオープンカーに乗り、旅する途中で出会う、様々な人間模様を描いたのがこのシリーズ。
(Get Your Kicks On) Route 66 はボビー・トゥループが1946年に作詞・作曲したポピュラー・ソングでジャズのスタンダード曲。
同年に、ナット・キング・コール盤、ビング・クロスビー＆アンドリュース・シスターズ盤がそれぞれヒットしたが、ジョージ・マハリスがテレビ・シリーズ「ルート66」の主題歌としてロックンロール風に歌ったところ大ヒットした。
